چکیده

A method is described for the study of transport processes in ghosts isolated from rat adipose cells. With this procedure it was possible to follow the time course of potassium uptake, potassium release, and sodium release. The uptake of potassium was stimulated by sodium and was inhibited by ouabain at concentrations down to at least 10e6 M. In the presence of pyruvate (1 mu), potassium uptake showed a more rapid initial increase, and the steady state level was more than doubled. Glucose (5 111~) produced a smaller stimulation of potassium uptake. Cyanide, 2,4-dinitrophenol, or incubation in the cold diminished both potassium uptake and content. Potassium efllux, unlike uptake, was not altered by ouabain or pyruvate; a half-life of 45 min was found. Sodium ion was exchanged but not accumulated by ghosts. Efflux studies showed that sodium was released with a half-life of 10 min. It is concluded that potassium is accumulated by ghosts of fat cells via a sodium-linked, energy-requiring process characteristic of an intact plasma membrane.

Effects of cytochalasin B, colchicine and vincristine on the metabolism of isolated fat-cells.

1. Colchicine and vincristine only slightly inhibit the metabolism of glucose to CO(2) and lipids by isolated fat-cells. 2. Prolonged incubation with these agents causes no further inhibition. 3. Cytochalasin B, however, inhibits glucose metabolism to both CO(2) and lipids in fat-cells. 4. However, at a concentration that causes a strong inhibition of glucose metabolism cytochalasin B is withou...

Response of trypsin-treated brown and white fat cells to hormones. Preferential inhibition of insulin action.

Insulin did not stimulate glucose metabolism or inhibit lipolysis in brown fat cells isolated by digestion of rat interscapular brown adipose tissue with collagenase plus 1 mg of trypsin per ml. Trypsin-treated cells responded normally to catecholamines and to the antilipolytic agent, prostaglandin El. The stimulation of respiration by lipolytic agents and free fatty acids was not affected by t...

Metabolism of Isolated Fat Cells I. PZ$‘E:CTS OF HORMOKES ON GLUCOSE MtiTABOLIShI ASD LIPOl,YSIS

Increased attention has been focused on the metabolism of adipose tissue and its marked sensitivity to various hormones (for reviews, see Vaughan (1) and Jeanrenaud (2)). In an attempt to obtain a homogeneous preparation of fat cells, it was found that if rat adipose tissue is t,reated with collagenase, fat cells are liberated. Owing to their high fat content, the fat cells can be separated fro...

Effects of insulin on glucose metabolism in isolated human fat cells

Isolated fat cells were used for the study of in vitro effects of insulin on glucose metabolism in human and rat adipose tissue. In human subcutaneous fat cells, effects of insulin could be detected at concentrations of glucose in the medium from 1 to 10 pmoles/ml. Cellular responsiveness was inversely proportional to the glucose level. At a constant concentration of 6 pmoles of glucose per ml,...

Multilineage Differentiation Activity by the Human Umbilical Vein-Derived Mesenchymal Stem Cells

Background: Mesenchymal stem cells (MSC) are a very promising transplantable stem cell source for a variety of cell replacement therapies. As the main source of MSC is bone marrow (BM), most of studies have been done on BM-derived MSC (BM-MSC). Umbilical cord (UC)-derived MSC (UC-MSC) which are recently introduced, is one of the good alternative source for these cells. The objective of this stu...
